Muscle contraction is attenuated twice, by blocking ion entry with Leuphasyl, and by modulating the formation of the SNARE complex with Argireline.

Muscles contract when they receive neurotransmitters released from a nerve cell and botulinum toxin type A paralyzes the muscle by selectively blocking the release of acetylcholine at the neuromuscular junction. BoNTA irreversibly fragments the SNAP-25 protein, preventing the formation of the SNARE complex. The nerve signal cannot be transmitted, so the muscle cannot contract. Argireline is an N-terminal analog of SNAP-25 and competes with the natural protein for a position in the SNARE complex, destabilizing its formation without breaking any of its components. If the SNARE complex is slightly destabilized, the vesicle cannot release neurotransmitters. Leuphasyl is a pentapeptide that mimics the natural pathway of enkephalins, acting on the formation of expression wrinkles in a new way. It is a modified enkephalin that binds to the enkephalin receptor on the outside of nerve cells.
