Active ingredients: Oligopeptide-68, Oligopeptide-58, Oligopeptide-51, Oligopeptide-34, Decapeptide-15.

BCN Lumen-Peptides is a sterile solution based on a combination of 3 types of active ingredients for the treatment of hormonal, aging and/or post-inflammatory hyperpigmentation. It acts at different levels of the metabolic process of melanin pigment formation in the skin, decreases melanogenic factors, inhibits melanin synthesis, increases cellular detoxification and reduces melanosome transfer to the keratinocyte.
Thanks to the exclusive new generation peptides it contains, it intervenes at the subcellular level by modulating the genetic transcription of key elements of melanogenesis. It also regulates intra- and intercellular communication ensuring the survival of the cells involved.
BCN Lumen-Peptides adds other ingredients such as hyaluronic acid, vitamins, amino acids, minerals etc. achieving a global anti-aging treatment for a clearer, brighter, hydrated and even skin tone.

Skin pigmentation is the result of melanin synthesis by melanocytes through a process called melanogenesis. Once synthesized, melanin pigments are transferred to keratinocytes (the most abundant cells on the surface of the epidermis), in vesicles called melanosomes. The more melanosomes reach the surface keratinocytes, the darker the skin becomes. The skin actually produces two types of melanin: pheomelanin in fair-skinned people and redheads, and eumelanin in brunettes and blacks.

Melanin synthesis is produced from the amino acid tyrosine. Tyrosine, in the presence of the enzyme tyrosinase and oxygen, is oxidized to dopa, which is in turn oxidized to dopaquinone. Dopaquinone then spontaneously changes to leucodopachrome, which is readily oxidized to dopachrome. The carboxyl is removed from the dopachrome and the dopachrome rearranges to melanin.

The combination of active ingredients of BCN Lumen Advanced-Peptides acts on all the phases of this melanin production process to achieve a depigmenting, whitening and skin-toning effect, thanks to the synergistic and combined action of three types of active ingredients: new generation biopeptides (oligopeptides 51, 34 and 58, decapeptide 15 and acetylglycyl beta-Alanine), hypopigmenting tyrosinase inhibitors (α-arbutin and licorice root extract) and antioxidants, such as ascorbic acid, thiocytic acid, vitamin B3 and glutathione.

This preparation also reduces melanogenic factors, inhibits melanin synthesis and increases cellular detoxification. These actions result in the following effects:

  • Reduction of melasma (excess pigmentation).
  • Reduction of age spots.
  • Reduction of hyperpigmentation in pregnancy.
  • Reduction of post-inflammatory hyperpigmentation.

The function and mechanism of action of each component are as follows:
Biopeptides:

Oligopeptide-51
, whose whitening effect is due to its inhibitory action at the level of the microphthalmia-associated transcription factor (MIFT) cellular pathway, which is the master gene for melanocyte survival and a key factor in regulating the transcription of melanin-generating proteins in the physiology of normal pigmented cells, such as tyrosinase-related proteins type 1 and 2 (TRP 1 and TRP 2). The result is the reduction of the transcription factor and in turn of tyrosinase. It also inhibits the expression of the melanogenesis marker and induces morphological changes in melanocytes.

Oligopeptide-34 also acts by inhibiting melanin synthesis by reducing both the MITF factor and the proteins related to tyrosinases 1 and 2. This peptide also acts at the melanosome level, reducing its quantity and blocking the transfer of pigment from the melanosome to the keratinocytes. Likewise, oligopeptide-34 has an anti-inflammatory effect, since on the one hand, it decreases the proliferation of activated T cells and reduces the volume of proinflammatory cytokines, as well as their level of transcription, translation and expression in case the inflammation is due to polysaccharide endotoxins. On the other hand, it decreases the translocation to the nucleus of the p65 subunit of the protein complex of the nuclear factor enhancer of activated B-cell kappa light chains (NF-kB), which plays a key role in inflammatory and immune responses. Finally, this peptide increases the expression of collagen and fibronectin with a consequent anti-wrinkle effect.

Oligopeptide-58
and Oligopeptide-68, like the previous peptides, inhibit the action (at the level of the MITF cellular pathway) and thus the transcription of the main melanogenic proteins, tyrosine kinases 1 and 2, as well as protein kinase C-β (PKC-β). Thus, it reduces melanin synthesis in the melanocyte. It also decreases the formation of dendrites or prolongations of the melanocyte, which allow the melanosome to reach the keratinocytes of the epidermis.

Decapeptide-15
has the same mechanism of action as the previous peptides.

Acetylglycyl beta-Alanine
is a peptide that inhibits the two cytokines that stimulate melanocyte generation:
stem cell factor and endothelin 1, which are activated by UV rays and modulate melanocyte response. It also inhibits the expression of the MITF gene, decreases intracellular melanin content, in addition to melanophilin, both at the messenger RNA and protein levels.

Non-peptide hypopigmentants enzymatic tyrosinase inhibitors:

Alpha-Arbutin is the glycosylated derivative of hydroquinone, which is the reference depigmentant used in dermatology, but with better tolerance, so it can be used in cosmetics. It exerts its depigmenting effect by effectively inhibiting tyrosinase activity and,
therefore, melanin production. It accelerates the breakdown and elimination of melanin. Due to structural similarities with tyrosinase substrates, arbutin acts by a competitive mechanism, binding to the active sites of the enzyme. In addition, it has the ability to inhibit DHICA polymerase (5,6-dihydroxy-indole-2-caerboxylic acid), a product resulting from the oxidation of tyrosine by tyrosinase, at
. It also inhibits the maturation of the enzyme.
It also inhibits the maturation of melanosomes, thus reversing in a decrease of melanin synthesis.

Glycyrrhiza Glabra root extract, which contains glabridin, a molecule of great structural similarity to hydroquinone, as well as liquiritin and
isoliquertin. It disperses melanin, influences melanogenesis and skin inflammation. Reduces dopa-positive melanocytes (dopapositive reaction occurs when dopa is added to tissue containing tyrosinase-containing melanocytes). It also has anti-inflammatory
(cortisol-like) and epithelializing properties.


Antioxidant active principles:

3-O-ethylascorbic acid, it is a more stable acid than Vitamin C. It has antioxidant action for reactive oxygen species (ROS) related
to melanogenesis. It inactivates tyrosinase by binding to the active site of the copper-containing enzyme. It also reduces oxidized dopaquinone.

Thiocytic acid or α-Lipoic acid, which in addition, is also a non-enzymatic inhibitor of tyrosinase. It acts by inhibiting the expression of MITF. Its capacity
antioxidant is the basis of its depigmenting action. It exerts both antioxidant and anti-inflammatory action.

Vitamin B3, niacin or nicotinamide, acts as the previous one, as an antioxidant and anti-inflammatory, prevents photoimmunosuppression and the passage of
melanosomes (containing melanin) from melanocytes to keratinocytes, so that melanin is not released in these epidermal cells.

Glutathione neutralizes free radicals, increases cellular detoxification and changes the production of dark eumelanin to light pheomelanin. It inhibits tyrosinase indirectly by binding to the active site of the copper-containing enzyme.

Vitamin E, by its strong antioxidant action, delays the aging of skin cells. Moreover, it fights free radicals that influence the deterioration of skin tissue and are responsible for the early appearance of wrinkles. In addition, it offers sun protection while
regenerates sun-damaged skin due to its regenerative and healing properties. It protects the skin from environmental toxins derived from pollution and aggressive weather conditions.
